ユビキチン・プロテアソームによる出芽酵母細胞機能制御に関する分子遺伝学的研究

研究概要	出芽酵母26Sプロテアソームの構成蛋白質:nin1-1変異体は26Sプロテアソームのサブユニットの一つをコードしている。nin1-1変異のマルチコピーサプレッサーを二種分離し、SUN1およびSUN2と命名した。SUN1はそのアミノ酸配列からマルチユビキチン鎖認識蛋白質であることが予想され、実際にその活性があることを証明した。この活性は、26Sプロテアソームが基質を認識するときに必須であると考えられているが、SUN1破壊株は野生株と同様の生育を示した。この事実は、26Sプアロテアソーム中にもう一つのマルチユビキチン鎖認識蛋白質があることを示唆している。SUN2は必須遺伝子であることが分かった。SUN2はヒト26プロテアソームのサブユニットp58およびショウジョウバエのDoxA2と機能的にも構造的にも高い相同性を示した。Sun1とSun2蛋白質は酵母の26Sプロテアソームのサブユニットであることも示した。ユビキチン経路の新規遺伝子の解析:温度感受性突然変異体保存株中の一つYTG1株はBUL1やRSP5およびUBI1などのユビキチン関連遺伝子によってサプレスされる。このことから、YTG1遺伝子もユビキチン経路に関連のある作用を持つ遺伝子であろうと推定されるが、その本体はまだ明らかになっていない。今回、ytg1/+二倍体株の遺伝子破壊を網羅的に行うことが可能なバンクを用いて、形質転換と同時に温度感受性になるものを選択し、その中から、ytg1の対立遺伝子の破壊株を得た。この破壊株から、挿入DNA断片を目印に染色体DNA断片をクローニングし、その塩基配列を決定した。YTG1がコードするアミノ酸配列を知ることが出来たが、この情報からはYtg1蛋白質の機能について新しい知見は得られなかった。今後、YTG1遺伝子と相互作用する遺伝子の検索などを通じてその働きを明らかにする予定である。
