研究概要	感染性プリオンを試験管内で複製することを最終目標として、試験管内で微量のプリオンと多量の正常プリオン蛋白の接触により正常プリオン蛋白の構造を変えることを目論んだ。以前、動物脳から正常プリオン蛋白を多量に精製することに失敗しているため、今回は組換プリオン蛋白を用いることを計画した。さらにプリオン蛋白の精製の困難さから、組換プリオン蛋白のN端にヒスチジンタグを結合し、キレートカラムでアフニティー精製を導入した。今回われわれの用いた系で、プリオンに見られるように、微量のプリオンを添加することにより組換プリオン蛋白のαヘリックス含量が減少し、βシート含量が増加すること、蛋白分解酵素抵抗性に変化すること、プリオン蛋白の一部に相当する合成ペプチドの添加により阻害されること、さらにこのように変化したプリオン蛋白を次の新たなプリオン蛋白に添加することにより、プリオン添加と同様の構造変化を引き起こすことを見出した。唯、この系では、蛋白分解酵素抵抗性に変わったプリオン蛋白の状態がプリオンと同様の構造を反映しているとはいえない可能性が示唆された。この結果、真のプリオン複製に至らなかったが、プリオン複製のために、プリオン蛋白以外の要因が必要か否かを解析するために適した系として使用できる可能性が示唆された。一方、本研究をサポートする周辺領域の研究として、プリオン蛋白検出法の改良、牛プリオン遺伝子の完全1次構造解析、発現調節、羊プリオン遺伝子多様の解析、さらに人アルツハイマー病の危険因子であるApoE蛋白遺伝子が、プリオン病の危険因子ともなる可能性について、羊スクレイピーでの検討等も行った。
