自律神経シナプス伝達に対するサイトカインの作用

研究概要	近年、脳の外傷や拘束ストレスの際にマクロファージや単核球等から遊離されるインターロイキン-1(IL-1)が脳内で産生、遊離され多くの神経作用をひき起こす。本研究は、自律神経系に対するストレスの影響について調べる目的で、ウシ蛙やラットの交感神経節や、副交感神経である骨盤内神経叢(pelvic plexus)、および自律神経系の上位中枢である大脳中隔核におけるシナプス伝達に対するIL-1βの作用について調べた。IL-1βはウシ蛙交感神経節細胞に約5mV程度の過分極電位を発生させたが、膜抵抗にはあまり変化がみられなかった。交感神経節の節前神経を電気刺激することによって発生した速い興奮性シナプス後電位(EPSP)はIL-1β(100pM)投与により51±5%(n=4)振幅が減少した。mEPSPの振幅も同様に抑制されたが、頻度には変化が見られなかった。ラットのpelvic plexusには静止電位が-55±6mV(n=24)、膜の入力抵抗48±9MΩ(n=43)で、外向き整流を示す細胞群と静止電位が-64±5mV(n=33)、膜の入力抵抗98±8MΩ(n=33)で、内向き整流を示す2種類の細胞群が存在している。前者に属する細胞でIL-1β(100pM)は5mV以下の過分電極位を発生した。海馬CA3ニューロンから中隔核への神経入力を通す海馬采に電気刺激を行うと興奮性シナプス後電位(EPSP)、抑制性シナプス後電位(IPSP)、緩徐過分極性シナプス後電位(LHP)が発生する。IL-1β(100pM)は静止電位には変化を起こさず、EPSPとLTPの振幅を2相性に変化させた。即ち始め抑制し、5-10分後に増強した。以上、本年度得られた実験結果をまとめると、末梢神経系では自律神経節細胞における過分極電位発生とEPSPの振幅の抑制、中枢神経系では、中隔核ニューロンにおけるEPSPとLHPの振幅の抑制が生じる。今後、これらの作用機序について検討し、細胞内シグナル伝達機構に関与について明らかにしていく。
