自律神経シナプス伝達に対するサイトカインの作用

研究概要

平成10年度の研究では、自律神経節におけるIL-1βの作用メカニズムについて細胞内微小電極法及びホールセルパッチクランプ法を用いて調べた。その結果、IL-Iβは自律神経節(交感神経節や骨盤神経叢)細胞において、過分極電位のみならずこれに先行して一過性の脱分極電位を発生することを見いだした。これらIL-1βの活性ドメインであるIL-1β 163-171は、IL-1βと同じ作用を持つことがわかり、このことはIL-1βの作用が特異的なものであることを意味している。今回、IL-1βによる脱分極電位や過分極電位はGABA誘起脱分極電位に性質が類似していることや、lL-1βによって発生した二相性電位がピクロトキシンによって阻止されることから、内因性のγ-アミノ酪酸(GABA)を遊離することによって生じている可能性が示唆された。特にラットのMPGにはGABAの結合部位や合成酵素が存在することが知られており、この可能性は高い。また、長い経過を持つ二相性電位は膜透過型のサイクリックGMPによっても発生することがわかり、IL-Iβがグアニル酸シクラーゼの活性化を介して過分極電位を発生している可能性が考えられた。今回はパッチクランプ法を用いた研究で、単離培養した交感神経節細胞におけるIL-1β誘起過分極電位がG-蛋白を介して発生している可能性も示唆されており、G-蛋白-グアニル酸シクラーゼ系の細胞内シグナル伝達機構が関与していることが窺える。また、自律神経節におけるコリン性シナプス伝達に対するIL-1βの作用は、シナプス前終末からのアセチルコリン放出を調節するいわゆるシナブス前調節であることも判明した。現在そのメカニズムについて検討中であるが、少なくともEPSPの増強作用にはサイクリックAMPが細胞内情報伝達系として働いているようである。一方、IL-1βによるアセチルコリン放出の抑制にはシナプス前終末のGABA_B受容体を介し、Ca^<2+>チャネルを抑制すると考えられる。
