研究概要

研究の背景と目的:現在の遺伝子工学によって産生されるリコンビナントタンパク質の中には、本来そのタンパク質に結合している糖鎖が欠落しているため、生理活性に問題のあるこのがある。本研究は、この問題を解決するため、プロテオグリカンをモデルとしエンド型グリコシダーゼの有する糖転移反応により、生理活性を有する糖鎖を合成し、これをリコンビナント体に導入する技術の確立を目標としている。
研究の実績:
1.糖鎖再構築法の改善と再構築された糖鎖の構造解析:精巣性ヒアルロニダーゼのオリゴ糖単位の糖転移活性を利用し、コンドロイチン硫酸系の糖鎖を、任意にデザインされた通りの配列にする方法を確立した。またその再構築された糖鎖の構造を質量分析装置により解析する道を拓いた。
2.再構築糖鎖をペプチドに導入するためのキャリヤ-の検索:プロテオグリカンのコアタンパク質と糖鎖の結合部位は、"キシロース-セリン"という構造を有しており、この構造が糖鎖をペプチドに導入するためのキャリヤ-となる。このキャリヤ-構造が培養したヒト皮膚線維芽細胞の培地から発見され、その検討が進められている。
今後の課題:1.グリコシダーゼの糖転移活性を利用した糖鎖の組み換え法の他の種類の糖鎖への適用を拡大すること。2.再構築糖鎖をペプチドに導入するためのキャリヤ-糖鎖を確立すること。及び再構築糖鎖をペプチド中に位置選択的に導入する方法を確立すること。
