ループス腎炎原性抗体の血管内皮細胞に対する新たな傷害機構の解析

研究概要	昨年度に引き続き、MRL/lprループスマウスから得た腎炎原性単クローン性抗体が血管内皮細胞の傷害を介して糸球体病変を発症する機構を解析し、新たに以下の成果を得た。 1。ループス腎炎原性抗体B1が血管内皮細胞に積極的なendocytosisを誘導せしめる機構:B1抗体は少なくとも血管内皮細胞膜成分に対する抗原結合活性を有さず、細胞のFcレセプターや酸化LDLレセプターを介さずにmicrofilamentの機能を介してendocytosisされ、transendothelial transportおよびリソソーム系にて処理されることが明らかとなった。臨床応用を目的として、reno-vascular diseaseを有する患者中の一部の患者血清lgGがendocytosisされることを見出し、新たな疾患カテゴリーの提唱、病理発生の解明の糸口を得た。 2。ループス腎炎原性抗体2B11.3が血管内皮細胞にE-selectinの発現を誘導する機構:2B11.3抗体によるE-selectinの発現誘導はTNFによる機序とは異なるもので、endocytosisがシグナルとして作用し、transcriptionレベルで誘導していることが判明した。正常マウスにこの抗体を投与すると糸球体内皮細胞にE-selectinが誘導され、糸球体腎炎が発症すること、soulible E-selectinトランスジェニックマウスでは腎炎発症が抑制されたことから、この抗体によるE-selecinの誘導そのものが、糸球体腎炎の発症のinitiationとなってことを明らかにした。
