マスト細胞におけるmi転写因子の機能とその異常の分子病理学的解析

研究概要	マウスのmi遺伝子座はbasic helix-loop-helix leucine zipper (bHLH-Zip)型転写因子(以下mi転写因子)をコードしている。mi突然変異遺伝子は、basic領域に列ぶ4個のアルギニンのうち1個が欠失しており、ホモのmi/miマウスではマスト細胞の数が正常(+/+)マウスの1/3に減少するとともに、その分化形質の異常を示す。我々はマスト細胞の減少が、マスト細胞の最も重要な増殖因子であるstem cell factor (SCF)の受容体であるc-kitの発現の低下のためであることを報告していたので、本年はmi転写因子がc-kitの転写に関与しているかどうかしらべた。mi/miマウスと対照の+/+マウスから培養マスト細胞を得て、c-kitのmRNA発現量をくらべるとmi/mi培養マスト細胞で有意に低い。このmi/mi培養マスト細胞に正常のmi転写因子をコードするcDNAを導入するとc-kit・mRNAの発現は正常化するが、アルギニン1個を欠失する異常のmi転写因子のcDNAを導入したのでは、c-kit・mRNAの発現は低下したままである。c-kit遺伝子の5'上流にはbHLH-Zip型転写因子により認識されるCANNTG配列が存在する。我々は正常のmi転写因子がこの配列を持つオリゴヌクレオチドに結合すること、さらにc-kit遺伝子のプロモーターをルシフェラーゼ遺伝子の上流につないだものと、正常のmi転写因子をコードするcDNAを同時にNIA/3T3細胞に導入すると、ルシフェラーゼの活性が有意に上昇することを示した。我々はすでにマスト細胞特異的プロテアーゼの1つマウス・マスト細胞・プロテアーゼ6(MMCP-6)の転写がmi/mi培養マスト細胞で著明に低下していることを報告していたが、本年度はMMCP-6遺伝子の発現もmi転写因子により実際に支配されていることを示した。
