マスト細胞におけるmi転写因子の機能とその異常の分子病理学的解析

研究概要	マウスのmi遺伝子座はbasic-helix-loop-helix-leucine zipper型の転写因子(以下MITF)をコードしている。MITFがマスト細胞の分化過程に影響することはmi/miミュータント・マウスの組織でマスト細胞が少なく、その分化形質が異常であることからも明らかである。本研究では、MITFによりその発現が影響を受ける遺伝子を、正常(+/+)マウス由来のマスト細胞とmi/miマウス世来のマスト細胞を比較することによって調べ、候補として得られた遺伝子については、その遺伝子のプロモーター領域にMITFなどbasic-helix-loop-helix-leucine zipper型転写因子が結合しうるCANNTG配列があるか、さらにその遺伝子のプロモーター領域をルシフェラーゼ遺伝子の上流に結合した際、ルシフェラーゼ遺伝子の転写を亢進させるかどうか調べた。nerve growth factor(NGF)の受容体の1種p75、マスト細胞プロテアーゼ(MMCP-4)、グランザイムB、セロトニン合成に重要な役割をはたすトリプトファン・ヒドロキシラーゼなどの上流にあるCATTGA配列にはMITFの結合がみられ、これら遺伝子のプロモーター領域をルシフェラーゼ遺伝子の上流につなぐと、MITF存在下でルシフェラーゼ遺伝子の転写の亢進が認められた。これらMITFにその転写が影響されると思われる遺伝子のうち、グランザイムBとトリプトファン・ヒドロキシラーゼのcDNAは、+/+マウスの培養マスト細胞から得たcDNAライブラリーからmi/miマウスより得たcDNAライブラリーをサブトラクションすることにより得られたものである。このサブトラクション・ライブラリーはその転写にMITFの影響を受ける遺伝子を探索するにあたって有用であることが実証された。
