研究概要	(1)既に作製したJak3欠損マウスのT細胞分化を解析した結果、胸腺および末消で自己反応性クローンが除去されておらず、Jak3は増殖シグナルだけでなく、T細胞のネガティブ選択に重要な役割を果たしていることが明らかになった。 (2)T細胞分化におけるTCRシグナル系を解析するため、CD3ε鎖に注目し、特異的な結合分子をクローニングした。CD3ε細胞内領域と会合する分子としてCAST(CD3ε-associatesignaransducer)を得た。CASTはCD3εの膜近傍部に特異的に結合し、ζ鎖には結合しない。CASTは活性化に伴ってチロシンリン酸化を受け、機能的には、dominantegative型のCASTを導入するとIL-2産生系のシグナルとしてのNFATの活性化が抑制されることから、IL-2産生へのTCRシグナルにおける重要な役割を果たすと考えられる。 (3)T細胞分化のアビディティーによる調節をTCRの側から解析するために、HY特異的TgマウスとCD3ζ欠損マウスを交配したマウスを解析することによって、TCR発現レベルが低くMHC/ペプチドは一定という条件におけるT細胞選択を調べた。TCRレベルが下がりTCRシグナルが低下すると、雄でみられたネガティブ選択はポジティブ選択に変化し、雌でみられるポジティブ選択は無選択へと変化した。これからTCRシグナルレベルが直接T細胞選択を制御していることが明らかになった。 (4)TCRβ鎖のみ発現する未熟胸腺細胞株として解析してきたKKFは、実はpTα-βダイマーを有することが判明した。そこでこの複合体を詳細に解析した結果、pTα-βダイマーとは、CD3γ,δ,εが会合するがζ鎖はほとんど会合しない。更に今回、この複合体中にカルネキシン及び我々の発見した12kd蛋白ダイマー(pTAC12)を同定した。その構造解析は未熟分化に重要なインサイトを提供すると思われる。
