研究概要	C型肝炎の細胞障害性Tリンパ球による肝細胞障害とセカンドシグナルの役割を免疫組織学的に検討したところ、C型肝炎ウイルスが感染すると肝細胞ではB7-1タンパクが誘導され炎症の程度と相関していたが、B7-2タンパクは発現していなかった。次に肝癌細胞株Hep3BにおけるHLAクラスI、B7-1、B7-2の発現をフローサイトメトリーで調べた。その結果、HLAクラスIは十分発現されていたが、B7-1とB7-2は発現が低レベルであった。このHep3B細胞にインターフェロンαとインタフェロンγを添加するとB7の発現が誘導された。Hep3B細胞はHLA-B35抗原を発現するため、HLA-B35遺伝子座を有するヒトから分離したリンパ球を、1)Hep3B細胞、2)インターフェロンαでB7-1を誘導したHep3B細胞、3)B7-1を遺伝子導入したHep3B細胞とともに培養したのち、Hep3B細胞に対する細胞障害性を検討した。その結果、E/T比が50/1で前2者の場合障害活性が5%、12%であったのに対し、B7-1遺伝子導入Hep3B細胞であらかじめ刺激したリンパ球は45%の障害活性を認めた。以上のことよりB7-1遺伝子導入肝癌細胞によるリンパ球の細胞障害活性の誘導が肝癌に対する治療に応用できる可能性が示唆されいた。
