増殖期消化管細胞の物理化学的刺激に対する応答性解析-物理集学的癌治療法の解析

研究概要	消化管細胞の物理化学的刺激に対する応答能を解析するために主に、家兎胃粘膜上皮細胞、線維芽細胞、平滑筋細胞の初代培養系を確立し、これを利用した胃粘膜損傷修復モデルを用いて各種の検討を行った。物理的刺激としては温度についてSV-40Large T antigenをtransfectしたラットより作成した胃粘膜上皮細胞株を用いた損傷修復モデルを用いて解析を行った。その結果によると33度の低温環境下では損傷の修復は主に細胞増殖により起こり39度の高温環境下では主に細胞により修復が惹起されることが明らかになった。また、37度環境下においては遊走と増殖により修復が起こることが明らかになった。化学的刺激としては、胆汁酸、アルコール、過酸化水素などを添加し修復の抑制が起こることを明らかにした。さらに、生物学的刺激として胃粘膜傷害及び胃発癌の重要な因子として注目されているHelicobacter pylori(Hp)の胃粘膜への傷害機序及び傷害修復機序への関与を解析するために初代培養胃粘膜細胞を用いた定量的損傷修復モデルを用いて解析した。cagA遺伝子、vacA遺伝子ともに陽性のHp株及び両遺伝子とも陰性のHp株より水抽出液を作成し初代培養胃粘膜細胞修復モデルに添加して、修復過程への影響を定量的に検討したところ両方の株で修復の遅延が惹起された事から新たな毒素の存在を示唆することができた。さらに、このHpにおける傷害惹起物質は血管内、および組織中マスト細胞の脱顆粒を引き起こしヒスタミンなどの炎症惹起物質を放出し、胃粘膜傷害の発生と炎症の引き金になる可能性を示唆する所見を得た。
