中枢ミエリン特異的遺伝子MOBPの基礎的・臨床的研究

研究概要

MOBP-/-マウスを作成し報告したが行動異常等は認めなかった。今回はこのマウスのradial component(RC)に注目し、解析した。RCは1960年代に発見された中枢ミエリンに特異的構造物でありその機能からintramyelinic tight junction(ITJ)とも呼ばれている。
MOBP-/-マウスと野性型マウスをパラホルムアルデヒド+グルタールアルデヒドで灌流固定後、視神経/脊髄を取り出し、タンニン酸を加えた液で浸漬固定し、電子顕微鏡でRCを観察した。また、ミエリン障害を引き起こすことで知られているヘキサクロロフェン(HCP)を経口投与し、同様に固定した後ミエリンの形態を観察した。野性型のマウスでは従来報告されているdiffuse typeのRCが観察されたが、MOBP-/-マウスではcondensed typeのRCが観察された。ノックアウトマウスでは、各RCの間隔は短く特徴的なジグザグ走行も失われている。HCP投与した野性型マウスでは従来報告されたVacuolationがintraperiod line(IPL)を割くように出現しているのが観察されたが、ノックアウトマウスではIPLに出現したVacuolation以外にmajor dense lineがゆるんだ像が観察された。
RCの構成成分は長い間謎であったが本研究によりMOBPがその構成分子の一つである可能性が示唆された。RCはITJとも呼ばれミエリンにおける一種のtight junctionとも考えられていたが、通常tight junctionを構成しているocculudinなどは発現していない。HCP投与実験からはMOBPがmajor dense lineのcompactionに重要な役割りを演じている可能性を示した。
