血管内皮細胞NO合成酵素の循環調節における意義-遺伝子工学的手法を用いて

研究概要	1.NO合成酵素の過剰発現マウスの作成生体内でのL-アルギニン-NOカスケードの役割、および種々の病態における意義を解明することを目的とし、内皮構成型NO合成酵素(NOS)の過剰発現マウス(トランスジェニックマウス)の作成を行なった。心血管系に特異的発現をもたらすと報告されているエンドセリン1-プロモーター(9.1kB)の下流にウシ大動脈内皮型NOS(beNOS)cDNA(4.1kb)をくみ込み、発生工学的手法を用いて3系統のトランスジェニックマウスを作成した。mRNAの発現はRNase protection assay法を用いて検討し、トランスジェニックマウスにおいてbeNOS特異的なmRNAを大動脈をはじめ、心、肺等主要臓器に認めた。NOS蛋白発現量の変化については、immunoblottingにて大動脈および肺で明らかに増加していることを確認した。免疫組織学的検討にて、この発現は血管内皮細胞に限局しており、しかも内因性マウス内皮型NOSと同じく細胞膜分画に認められた。この発現増加と一致して、3H-arginineから3H-citrullineへの変換にて測定したNOS活性も大動脈膜分画にてCa^<2+>依存性に約9倍増加していた。 2.NO合成酵素過剰発現マウスにおける血管反応性の検討 NOS過剰発現マウスの胸部大動脈の等尺性張力を測定し、内皮依存性血管弛緩反応の程度をコントロールマウスと比較した。acetylcholine,ATPγSによる内皮依存性血管弛緩反応は、予想に反しbeNOS過剰発現マウスにおいて明らかに減弱していた。現在さらに種々の血管作動物質に対する血管反応を検討中である。
