細胞周期進行における血管内皮および平滑筋細胞のイオン動態の制御機構とその破綻

研究概要	動脈硬化病変部では、血管平滑筋細胞の増殖異常が観察され、同時に、血管平滑筋細胞の緊張異常(血管攣縮、狭心症、高血圧症など)を伴うことが多い。本研究では、血管平滑筋細胞の増殖と血管機能との関係について、検討した。初年度は、血管平滑筋細胞のCaチャネルのタイプが細胞周期の進行に伴って、変化することを見出した(Circ.Res.79,1996)。さらに、本年度は、動脈硬化の進展に重要な役割を果たしていることが知られている血小板由来増殖因子(PDGF)の血管増殖作用と細胞内Ca^<2+>動態との関係について検討し、以下の結果を得た。血管平滑筋へのCa^<2+>流入を阻害するCa^<2+>拮抗薬は、PDGFによる細胞質Ca^<2+>濃度上昇と細胞周期進行作用を共に抑制した。しかしながら、Ca^<2+>拮抗薬が細胞質Ca^<2+>濃度を低下させる程度と細胞周期進行の抑制の程度は、必ずしも、相関しなかった。また、3種類のCa^<2+>拮抗薬の間において、細胞質Ca^<2+>濃度低下と増殖抑制に対するpotencyも相関が認められなかった。従って、Ca^<2+>拮抗薬は、細胞質Ca^<2+>濃度の低下以外のメカニズムによって、血管平滑筋の細胞増殖を抑制しているものと思われた。次に、初代培養血管平滑筋細胞を用いた実験により、PDGFによる細胞周期進行がチロシンキナーゼ阻害薬によって抑制されることが解ったので、in vivoの動物を用いた実験によって、チロシンキナーゼ阻害薬の効果について検討し、以下の結果を得た。長時間のPDGF刺激による豚冠動脈の狭窄形成を冠動脈造影法で検討した。チロシンキナーゼは、PDGF刺激による豚冠動脈の狭窄形成を著名に阻害することが解った。
