子宮筋腫の腫瘍性格に関する包括的研究

研究概要

子宮筋腫と子宮筋肉腫との違いをWestern blotting法で検討した結果、子宮筋腫には、細胞周期調節因子であるcyclinE、p34cdc2,p33cdk2、および発癌遺伝子c-rafなどの異常発現があり、その発現異常の程度は子宮肉腫により近いものであった。このことは子宮筋腫を良性腫瘍としてとどめるために一方で強力な細胞増殖抑制因子が作用している可能性を示唆した。また子宮筋腫には、p16,p21,p53などの腫瘍抑制因子の異常発現が見られる事より、筋腫では、細胞周期調節因子や発癌遺伝子の異常発現があるものの、腫瘍抑制遺伝子の発現により、より正常に近い機能を持つことによって良性腫瘍としての性格を維持しているのではないかと考えられた。また子宮筋腫、子宮筋、子宮平滑筋肉腫との違いを、differential display法で解析し結果、ポリAテ-ルに相補的なプライマー3種とランダムプライマー8種の組み合わせにより24通りのDifferential Displayを行い、正常平滑筋特異的、平滑筋肉腫特異的と思われる遺伝子10数個を単離した。子宮筋腫に特異的に発現する遺伝子として単離された遺伝子の一つはtropomyosinと相同性が高く、また子宮平滑筋肉腫に特異的なHG-3fは、塩基配列解析の結果、大腸菌DinGと相同性のある遺伝子であることが分かった(human DinG)。tropomyosinとhuman DinGの発現の特異性を確認するためにRT-PCR法を行ったところ、tropomyosinは子宮筋腫に、human DinGは平滑筋肉腫に有意に強く発現していた。human DinGは、DNA helicase様構造を持ち、DNA合成に直接関与することが推察されることから、human DinGの過剰発現が子宮平滑筋肉腫の悪性化に関与する可能性が示唆された。
