歯根膜神経の免疫細胞化学的、神経生物学的研究:発生学的基盤と実験モデルによる検討

研究概要

1.ラット切歯歯根膜に密に分布する機械受容器であるルフィニ神経終末の生後発育過程をすべての神経線維が染色可能なprotein gene product 9.5(PGP)抗体を用いた免疫電顕法にて検討した。その結果、生後1日目ではルフィニ神経終末は無随無鞘の軸索として観察され、4日目になると複数のシュワン鞘でおおわれた軸索終末が観察された。この軸索終末内には無数の小胞様構造とミトコンドリアが含まれていた。経日的にミトコンドリアの数は増し、その大きさも大きくなっていった。切歯が機能咬合を開始する生後15日目に初めて軸索終末から伸び出る指状の突起が観察され、また軸索終末周囲をおおう基底膜様構造も肥厚化していった。機能咬合が完成される生後25-26日になると、成体で観察されるような微細構造学的特徴をもつ歯根膜ルフィニ神経終末が観察され、歯根膜ルフィニ神経終末が観察され、歯根膜ルフィニ神経終末の成熟には機能刺激の付与が重要であることが示唆された。
2.ラット臼歯に実験的に咬合性外傷を付与し、その際の歯根膜ルフィニ神経終末の形態変化をPGPの免疫細胞化学で検討した。咬合性外傷の様な外来刺激にルフィニ神経終末は容易に反応した。実験開始後2日でに歯根膜ルフィニ神経終末は膨化しその外形は不鮮明となり、電顕下では巨大な原形質突起が周囲歯根膜組織に伸びだしていた。ラット臼歯歯根膜が咬合性外傷刺激に順応した7日になるとその形態は実験群と同じになった。以上の観察結果は咬合性外傷のような外来刺激により、歯根膜ルフィニ神経終末が容易に形態変化を引き起こすことを示している。
本研究の遂行には科学研究費補助金により購入した超ミクロトームが活用された。
