歯根膜神経の免疫細胞化学的、神経生物学的研究:発生学的基礎と実験モデルによる検討

研究概要

1.実験的に歯を移動させ、神経再生・発生時に一過性に発現し、重要な役割を果すgrowth associated pritein-43の歯根膜Ruffini神経終末での局在の変化を検討した。正常状態ではGAP-43の免疫反応はシュワン鞘に限局していたが、、移動に伴う組織改造が活発な直に対応して、GAP-43の免疫反応が通常は免疫陰性である軸索終末に出現し、また、終末シュワン細胞細胞質に出現した。しかし、組織改造が元のレベルに戻ると、消失することが示された。
2.機械受容機構に重要な役割を果たしているカルシウムイオンと高い親和性で結合するカルシウム結合タンパクの歯根膜神経における分布を検討し、歯根膜Ruffini神経終末がcalbindin D28Kとcalretininを含むことを明らかにした。
3.歯根膜ルフィニ神経終末の軸索終末細胞質にcarbonic anhydrase isozymeIIが含まれていることを免疫細胞化学的手法により明らかにした。
4.交感神経系の伝達物質であるノルアドレナリンと共存していることが知られるneuropeptide Yが歯根膜Ruffini神経終末の再生過程の際に一過性に出現することを免疫電顕法を用いて明らかにした。
5.化学的消化法を用いて歯根膜Rluffini神経終末の立体構造を明らかにし、歯根膜Ruffini神経終末に付随しているシュワン細胞の突起が歯根膜Ruffini神経終末の位置の維持・固定に重要な役割を果たしていることが明らかにされた。
これら一連の研究の遂行には本科学研究費補助金により購入した超ミクロトームが活用された。
