研究概要	最近、研究代表者らは、植物の消化酵素として知られるヒト膵液Phospholipase A_2-I (hPLA_2-I)が膵癌細胞の増殖を促進する現像を発見した。さらに、膵癌細胞にはhPLA_2-Iに対する得異的レセプターの存在することを明らかにし、その物理化学的諸性質を調べてきた。本研究では,hPLA_2-Iによって誘導される膵癌細胞(MIAPaCa-2)増殖促進作用が、hPLA_2-Iの消化酵素活性を介して行われるものであるかどうか調査した。その結果、hPLA_2-Iがその特異的レセプターに結合する際にはCA^<2+>を要求しなかった。また、hPLA_2-Iによる酵素分解産物であるリゾリン脂質には膵癌細胞の増殖促進作用が認められなかった。従って、この増殖促進作用はhPLA_2-Iの消化酵素としての活性を介する反応ではなく、hPLA_2-Iの特異的レセプターを介する応答であると結論した。 hPLA_2-Iが消酵素として機能するためには、膵臓で作られた前駆体型hPLA_2-IのN-末端アミノ酸が十二指腸中に存在するトリプシンの触媒作用により切断される必要がある。この切断されたものを成熟型hPLA_2-Iと呼んでいる。研究代表者らは、hPLA_2-Iによる膵癌細胞増殖促進作用は、成熟型hPLA_2-Iに特異的であることから、前駆体型および成熟型hPLA_2-Iにおける三次元構造を比較することを考えた。そのため、この2つのタイプのhPLA_2-Iを大腸菌で大量に産生させる系の確立を目指し、前駆体型あるいは成熟型hPLA_2-I遺伝子の構造遺伝子部分をそれぞれtrpプロモーターに連結した後、大腸菌に導入した。その結果、Lonプロテアーゼ欠損大腸菌変異株KY1436を宿主とした場合に、成熟型hPLA_2-Iの高発現が認められた。現在、精製したこの蛋白質を用いて結晶化の条件を調査している。さらに、引き続き、前駆体型hPLA_2-Iの大腸菌における大量生産系の確立も試みている。
