新規薬物動態研究モデルシステムの構築

研究概要	新規医薬品の開発段階で、また医薬品の適正使用を図る上で、薬物動態解析法の確立とin vivoを良く反映する代謝研究モデルシステムの構築が強く求められている。そこで、前年度の成果を基に、以下の検討を行った。まず、ESI-MSによる中性化合物の高感度測定法の開発を目指し、デヒドロエビアンドロステロンをモデル基質として固定化リコンビナントスルホトランスフェラーゼ用いる生理的条件下でのサルフェートへの誘導に検討を加えた。その結果、定量的にサルフェートへ変換されるばかりか、ESI-MSにて分子量に相当する特徴的な負イオンを生成し、本導体化法とLC/ESI-MSとの組み合わせが、水酸基を持つ中性化合物の高感度分析に有用なことが判かった。一方、グルクロン酸抱合を解析するうえに、グルクロニドの体内導態を把握することが重要となる。そこで、LC/ESI-MSを用い尿中胆汁酸3-グルクロニドの直接一斉分析を行い、病態時には、それらの排泄量が極めて低いことを明らかにした。また、肝ミクロソーム画分においては3位水酸基を介したエーテル型グルクロニドではなく、24位カルボキシル基を介したエステル型グルクロニドが優先的に生成することを実証し、胆汁酸のグルクロン酸抱合に新たな知見を加えることができた。引き続き、これらの知見を基に固定化薬物代謝酵素群を用いる代謝研究システムの構築を目的として、人工脂質膜にラット肝ミクロソーム画分を固定化し、エストラジオールをモデル基質として2位水酸化酵素活性に検討を加えた。その結果、酵素活性は単位体積当たりの固定化タンパク量の減少とともに著しく上昇し、また、固定化酵素をミニカラムに充填して用いるフローシステムでは、連続使用に伴う酵素活性の低下が、空気の注入によって回復されることが判明した。現在、グルクロニルトランスフェラーゼ活性に検討を加えており、これらの成果は、薬物代謝研究システムとして大きく役立つものと期待される。
