ヘモグロビン異常症の遺伝子治療に関する多面的アプローチ

研究概要	1. 遺伝子導入系の開発 AAVベクターを用いて、赤芽球系培養細胞を対象に発現効率の高い、加えて挿入部位によらない一定した発現系の確立を行った。前年度の結果からグロビン遺伝子の組織及び時期特異的な発現に関与するLCRを構成する3箇所のDNaseI超高感受性部位との組み合わせでは挿入部位に依存した発現が得られないことが分かったため、ニワトリのβグロビン遺伝子群で同定されたインスレータのコア配列を2個タンデムにつなぎ、これでHS2に連結したβグロビン遺伝子を囲み、組換えAAVを作成した。これを用いてクローンを単離し、その中でプロウイルスゲノムがインタクトなクローンを5個単離しグロビン遺伝子の発現を検討した。インスレータを持たないHS2では発現がクローン間で6.1%から172.1%と広がりがあった。一方インスレータを組換えたウイルス由来のクローンでは発現が52.8%から58.3%で、発現の平均は56.2%と挿入部位によらない発現が見られた。以上からこのインスレータを用いた組換えAAVベクターは遺伝子治療に有効な系であると結論した。 2. 分子病態の修復点変異(HbE変異,IVS-2内654位変異)が来す選択スプライシングのアンチセンスオリゴによる抑制効果をin vivoで検討するために、正常および各変異グロビン遺伝子をHeLa細胞にトランスフェクションして得られたステープルトランスフォーマントを用いて検討した。0.4μMの濃度の18塩基のアンチセンス-2'-0-メチルオリゴリボヌクレオチドをリポフェクション法で導入後、36時間まで30%程度の異常スプライシングの低下を認めた。またHbE変異による異常スプライシングを抑制するために、スプライシングの際ドナー部位の選択に関わっているといわれているSF2/ASFを強制発現させることにより検討した。前年度はトランジエント発現の系を行ったが、今回はステーブルな発現細胞を単離し検討した。4個の外来性SF2/ASFを発現しているクローンの解析から、SF2/ASFの転写量と異常スプライシングの抑制効果とが相関しており、内因性のSF2/ASFの1.46倍の量で抑制効果が約20%であることが明らかになった。
