研究概要

中枢神経特異的なプロテオグリカン型チロシンホスファターゼであるPTPζの脳形成における役割を明らかにするため、以下の研究を行った。
(1) PTPζの細胞内基質の探索
PTPζの細胞内基質分子を同定することを目的として酵母、two-hybrid systemを用いて、PTPζの細胞内領域と結合する分子をラット脳cDNAライブラリーから単離することを試みた。その結果、PSD-95ファミリーに属する分子群が単離された。またPTPζが中枢のシナプス部にPSD-95と共に極在することが示された。この知見はPTPζがシナプス後領域でシナプス可塑性の調節に関与することを示唆している。
(2) PTPζ遺伝子ノックアウトマウスの解析
PTPζノックアウトマウスの脳構造、行動解析、海馬におけるLTP、小脳におけるLTP、薬物に対する反応性等におけるMTとの違いについて検討した。その結果、行動、LTP、脳形成、薬物反応性において異なるprototypeを示すことが判明した。現在詳細な解析を行っている。
