研究概要	研究代表者は、これまでにNa/K-ATPaseのサブユニット会合ドメイン、阻害剤結合ドメイン、KやNaに感受性のあるドメイン(イオンセンサー)を同定した。本研究(平成8-9年の2年間)は、「イオンセンサー領域はATPase分子内の"あるドメイン"にイオン特異的に直接作用を及ぼし酵素活性を調節する」という分子論的作業仮説に対する直接的証拠を得ることを目的とする。本初年度は以下のような結果を得た。(1)N-末端(Met1-Leu69)に存在するNa-感受性部位(Na-センサー)によるATPase活性調節の分子機序を解明するため、Na-センサーをグルタチオン-S-メチルトランスフェーゼ(GST)との融合タンパク質として大腸菌で発現させ、グルタチオン-カラム法で精製した。単離・精製したNa-センサーペプチドをNa-センサーを持たないP-型ATPase(例えばCa-ATPase)のアッセイ系に加えると、ATPase活性は濃度依存性に上昇した。即ち、Na-センサーはATPase分子内の"あるドメイン"にイオン特異的に直接作用を及ぼし酵素活性を調節する(図書1)。(2)研究代表者は既に原子間力顕微鏡(AFM)を用いてNa/K-ATPase分子の形態を数オングストロームの解像力で調べ、分子中央にpore構造が存在するとを報告した。本年度は、この方法を定量的解析に用いるための手法を開拓した。即ち、AFMを用いてF^oF^1-ATPaseのF^0(H-ポンプ)においてもpare構造が存在することを示し、更に、F^0のサブユニット構造も提唱した(論文3)、また、DNA-タンパク質複合体を用いてAFMの定量的解析における作用性を確立した。タンパク質の大きさの決定、重合度の検討、特異的高次構造の検出等にAFMは有効であることが明らかになった(論文4と5)。
