ゲノミック・インプリンティングの生物学的意義

研究概要	哺乳類の個体発生、成長、行動には、父親、母親由来のゲノムは機能的に異なる役割を果たしている。このゲノムインプリンティングと呼ばれる現象の分子的原因はインプリンティング遺伝子と呼ばれる2種類の遺伝子群Paterma11y expressed genes(Peg)とMatema11y expressed genes(Meg)の存在によっていることを、これらPeg、Meg遺伝子群の体系的な分離によって、ほぼ実証できた。人工的に作製した母親由来のゲノムのみをもつ雌性単為発生胚や、父親由来のゲノムのみをもつ雄性発生胚と、正常受精胚の間での遺伝子サブトラクションは、新しく開発したPCRを用いた極微量の生物材料からの遺伝子サブトラクション技術により可能になり、これまでに8つのPeg遺伝子(Peg1-8)と5つのMeg遺伝子(Meg1-5)の分離に成功した。これらの遺伝子はマウス染色体上の7ケ所のインプリンティング領域にマッピングされることから、ゲノムインプリンティング現象が、PegとMegの2群の遺伝子の発現の過剰または欠除によっておこると考えることができるようになった。このうち我々が新規に同定したインプリンティング領域は5ケ所にのぼり、この現象の全貌を解析するための多くの知見を得ることができた。また、これらの遺伝子群の発現様式、発現部位を体系的に解析することにより、インプリンティング遺伝子の母親の卵成熟過程における刷込みの実体の解明、インプリンティング遺伝子が胎盤で共通に発現することから、この現象の生物学的意義に関しての提唱している新胎盤仮説など多くの業績をあげることができた。この研究から、胎盤形成という問題を通して、ゲノムインプリンテイングと哺乳類の誕生という生物進化の問題解明の糸口が掴めたと考えている。
