研究概要

核-細胞質間物質輸送機構の解明は、細胞増殖、細胞死、細胞分化など、様々な細胞機能に密接に関わる重要な研究テーマである。本研究は、細胞外からの増殖因子やホルモンの刺激に応答してシグナルとなる分子がどのようにして細胞質から核内に移行して、細胞外の情報を核に伝えるのかを分子レベルで明らかにすることを目的とし、インターフェロン・ガンマ刺激によって細胞質から核内に移行するSTAT1分子に着目して、以下に示す成果を挙げてきた。
1.STAT1分子は、その変異蛋白質を用いた解析から、従来より知られている、一次構造上、塩基性アミノ酸のクラスターからなるような核局在化シグナルを有さないことがわかってきた。
2.STAT1分子の細胞質から核膜孔までの輸送は、SV40T抗原の核局在化シグナルを持つ蛋白質と同様に、核膜孔ターゲティング複合体を形成することによって行われることがわかった。さらに、核膜孔ターゲティング複合体を構成する58kDa因子に関しては、ヒトの場合、現在、大きく2つのクラスに分類されているが、STAT1は、そのうちの1つであるNPI-1と呼ばれる分子でのみ運ばれることもわかった。
3.STAT1分子の核膜孔通過のステップは、SV40T抗原の核局在化シグナルを持つ蛋白質の場合と同様に、低分子量G蛋白質によるGTPの加水分解が必要であることがわかった。
以上のように、研究目的に沿って、STAT1の細胞質から核への移行のメカニズムの分子レベルでの解明が進んだ。
