転写因子STAT1をモデル分子とした核-細胞質間分子輸送制御のメカニズム

研究概要

核-細胞質間のシグナル伝達機構の解明は、細胞増殖、分化、細胞死など、様々な細胞機能に密接に関わる細胞生物学上の重要なテーマである。本研究では、インターフェロンなどのサイトカインの刺激によって細胞質から核へ移行して働く転写因子であるSTAT1分子に着目し、その細胞質と核の間の輸送に関わる因子の同定を通して、STAT1輸送のメカニズム解明を目指した研究を進め、以下のような成果を得た。
1.STAT1の細胞質から核への移行には、ヒトの場合、PTAC58(importin-α)ファミリーに属する分子の1つであるNPI-1によって特異的に認識されることが必要であることを明らかにしてきたが、その認識には、NPI-1分子のC末端領域が必要であることがわかった。この領域は、NPI-1分子が持つアルマジロ構造と呼ばれる、SV40T抗原などの核局在化シグナル認識に必要な特徴的領域とは異なり、NPI-1分子が2つの異なる核蛋白質を同時に結合して核へ輸送する可能性を示唆する結果となった。
2.STAT1を中心に形成されるNPI-1,PTAC97を含む複合体が、STAT1結合DNA配列と反応させることにより解離することが明らかとなり、STAT1が実際に核内で働くために必要と思われるNPI-1からの解離のメカニズム解明へ向けた研究成果を挙げることができた。
3.STAT1分子を核内にマイクロインジェクションし、その後の挙動を観察した結果、STAT1は温度に依存して能動的に核から細胞質へ遊出されるが、そのタイムコースは、ロイシンに富んだ核外輸送シグナルを持つ蛋白質の移行に比べてかなり遅いことがわかった。
