トランスジーン組込み部位に特徴的なDNA構造の解析と高頻度組換ベクターの開発

研究概要

形質転換植物の報告例は多いが、組み込まれたトランスジーンの発現制御を促すシスエレメントや組み込み自体に影響を及ぼすシスエレメントの研究例は少ない。申請者らはパーティクルガン法で得た形質転換細胞のトランスジーンが組み込まれたホスト植物染色体DNA近辺すなわちジャンクション領域の徹底的な構造解析を進める中で、ジャンクション部分のホスト側DNAはいわゆるNuclear Scaffold Attachment Region (S/MAR)と呼ばれるシスエレメントであることを初めて見いだし、この成果を手がかりにこの様なジャンクションS/MARがトランスジーンの組み込み自体に影響を及ぼすとの作業仮説のもとに研究を進めてきた。本年度は次の成果を挙げた。
1)従来のタバコ培養細胞に加えて、シロイヌナズナ植物の形質転換個体3系統のジャンクション部位の塩基配列解析を進め、その結果、シロイヌナズナ植物のジャンクション領域のホスト側DNAもS/MARを持っていることを初めて明らかにした。
2)タバコ細胞の他の形質転換系統でのジャンクションS/MARライブラリーの作成について研究し問題点を解明、解決した。
3)シスエレメントがExpression効率またはIntegration効率に及ぼす効果を明確に区別する目的で、GUS遺伝子を用いた選択培地を用いない形質転換細胞検出システムの構築について系統的に問題点を研究した。このシステムはまもなく完成し、これまでに得られているDNA断片やS/MAR配列の組み込みに対する効果を系統的に研究し、組み込みを促進するベクターの開発を成功させる計画である。
