研究概要	1.IP_3受容体の発現をβ-galactosidase活性で観察できるトランスジェニックマウスを開発して、中枢神経系における発現局在を解析した。これを用いることで、変動する発現パターンの解析が容易となった。 2.IP_3受容体のIP_3結合領域を大腸菌で発現させて、そのIP_3リガンド結合活性に関してアミノ酸レベルで解析した。N末端側の225〜578アミノ酸はIP_3結合の「コア領域」を形成し、この中でArg-265、Lys-508、Arg-511は必須の塩基性アミノ酸であことが解った。一方、N末端から224アミノ酸まではIP_3結合にむしろ阻害的に働くことを明らかにした。 3.バキュロウイルスベクター/昆虫細胞系で、機能的なIP_3結合活性を持つヒトIP_3受容体を大量に発現させることに成功した。発現したIP_3受容体のIP_3結合親和性はCa^<2+>の影響を受け、タイプ1IP_3受容体とタイプ3IP_3受容体では全く逆のCa^<2+>濃度依存性を示した。タイプ1は、10nM以下の低濃度のCa^<2+>で高親和性で数μMに上昇すると低親和性に至る、EC_<50>が100nM Ca^<2+>のシグモイドカーブを示した。逆に、タイプ3は、100nM付近からそれ以下のCa^<2+>では低親和性で1μM Ca^<2+>以上になると高親和性に達する、EC_<50>が872nM Ca^<2+>のシグモイドカーブを示した。いずれのタイプのCa^<2+>依存性の親和性変化は可逆的であった。従って、細胞内のCa^<2+>濃度とIP_3濃度の両方がタイプ特異的なIP_3誘導Ca^<2+>放出を巧妙に制御することが予想される。 4.IP_3受容体の膜貫通領域を含むC末端部分とGreen Fluorescent Protein(GFP)とを融合したタンパク質をアフリカツメガエル卵母細胞で発現させた。共焦点レーザー蛍光顕微鏡で観察すると、発現タンパク質は網目状に分布する細胞内膜(小胞体など)に存在し、核膜外膜にも多く存在した。これらは何れもCa^<2+>ストア膜であることからIP_3受容体本来の局在部位に合致する。また、クロスリンク実験で発現タンパク質が四量体を形成することが解った。以上のことから、IP_3受容体の四量体形成とCa^<2+>ストア膜への局在にはチャネル領域を含むC末端部分が重要であることが明らかとなった。
