ヒトレトロウイルスの病原性の解析と治療に関する研究

研究概要	本研究では我々が開発したHTLV-I感染症モデル動物(HAMラット、HTLV-Iトランスジェニックラット)を用いてその治療法を確立することを最終目的としている。あおのためにはその病態を正しく理解することが必要で本年度はそれを主に行い以下の成績を得た。 1)HAMラット病変脊髄のHTLV-Iの感染細胞の同定をin situ hybridazation法で行いミクログリオア/マクロファージ系細胞であることが証明された。経時的にはPCR法によりMT2投与後約3ヶ月頃より脊髄でのHTLV-I感染が証明された。競合的RT-PCRによるpXの発現は乏突起細胞のアポートシスが出現するMT2投与後約7ヶ月から急激に発現し15〜18ヶ月へかけてその発現量は徐々に低下した。また、TNF-αの発現も約7ヶ月から観察され、徐々に増加し11ヶ月移行比較的高いレベルで経過した。一方、アポトーシス抑制分子であるbcl-2発現は7-12ヶ月にかけ強く抑制されていた。 2)HTLV-Iトランスジェニックラット a)env-pXラット T細胞表面分子の解析により本ラットでは疾患発症以前からCD80/86やICAM-1などのco-stimulatory分子を発現した末梢T細胞が多く、導入遺伝子により末梢T細胞が前活性状態にある可能性が示唆された。env-pXトランスジェニックラットと正常同系ラット間での骨髄細胞移植では、唾液線炎、皮膚炎は正常ラットに移植可能であったが関節炎などほかの疾患は正常ラットに移入されなかった。一方、正常骨髄細胞で骨髄が再構築されたenv-pXラットでは唾液線炎、皮膚炎は完全に抑制されたがそれ以外の関節炎などは程度は軽いものの発症し、env-pXラットに発症する膠原病には少なくとも2通りの発症機序が関与していると考えられた。 b)Ick-pKラット Ick-pXラットが樹立できた。これらのラットの内いくつかの系統で胸腺腫の発生を確認したが、今のところ白血病の発症は確認されていない。
