合成ペプチドライブラリを利用したBRMの分子設計

研究概要

細胞死受容体のひとつであるFas抗原を介したアポトーシスに対するアンタゴニストの分子設計を試みた。
1.Fas抗原に対する作動因子であるFasリガンド(ヒト由来、281アミノ酸残基)の細胞外領域(179アミノ酸残基)の一次構造に基づいて、7〜14残基の長さのペプチドで構成される合成ペプチドライブラリーを作成した。
2.Fas抗原に対してアゴニスト作用をもつ単クローン抗体によるアポトーシス誘導系に作成したペプチドライブラリーを導入し、アンタゴニスト作用をもつペプチドを選別した。Fasリガンドの細胞外領域の中央部分に相当する2種類のペプチドにアポトーシス抑制活性が見いだされた。
3.得られた2つのペプチドに対して、アセチル基、ジニトロフェニル基、ベンジル基、フルオレニルメチルオキシカルボニル基など種々の化学修飾を施し、Fas介在アポトーシスに対するアンタゴニストの強さを調べた。その結果、Fasリガンドのジスルフィド結合で形成されるループに含まれる領域の12残基のペプチドにジニトロフェニル基を導入したものが有力候補になった。現在、ペプチドの伸縮、アミノ酸残基の置換を行い、より強力なアンタゴニストの分子設計を目指している。
一方、ランダムなアミノ酸配列に基づくペプチドライブラリー(5アミノ酸残基)を作成し、培養細胞にアポトーシスを誘導するペプチドのスクリーニングを試みた。スクリーニングは、アポトーシス誘導に伴う細胞内プロテアーゼの活性化をイムノブロット法で検出する方法を採用した。ペプチドの細胞内への導入法、受容体との親和性増強のための化学修飾法を検討している。
