合成ペプチドライブラリを利用したBRMの分子設計

研究概要	細菌感染と宿主細胞応答の関係を理解するために生体応答調節薬(BRM)の分子設計を行った。最も基本的な細胞機能のひとつである細胞死(アポトーシス)を取り上げ、Fas抗原を介して誘導されるアポトーシスに対する遮断剤の合成を試みた。すなわち、Fas抗原を活性化して細胞にアポトーシスを引き起こす分子、Fasリガンドの細胞外領域の一次構造に基づいて遮断剤の骨格となるペプチド部分の設計を行い、さらにペプチドに種々の化学修飾を施し、強力な遮断剤として完成させた。Fasリガンド由来の12アミノ酸残基のペプチド(アミノ酸残基番号215-226)のアミノ末端をジニトロフェニル基やトリフェニルメチル基で修飾すると、細胞傷害性抗Fas抗体によるヒトT細胞株Jurkatのアポトーシスを抑制した。このペプチド性アポトーシス遮断剤は、細胞をホルボールエステル/イオノマイシンで処理したときに引き起こされるアポトーシスに対しても抑制効果があった。すなわち、T細胞受容体を介して誘導される天然のFasリガンドに対する遮断作用が示されたわけである。これらのアゴニストは、細胞表層のFasに対して修飾ペプチドと競合せず、リガンド(抗体)-受容体複合体に結合したペプチドがFasの構造変化あるいはFasから細胞質への情報伝達を抑制していると考えられる。本研究で確立したペプチドの化学修飾法を利用して、さらに2つのアポトーシス遮断剤が得られた。興味あることに、どの遮断剤のペプチド部分もFas・Fasリガンドの接触面と考えられる領域に位置していた。現在、これら遮断剤の作用機構の解析を進めており、個体レベルでの実験も予定している。
