研究概要	本年度は、特定のHLAクラスII分子により提示されたペプチドに特異的なヒトT細胞クローンと、抗原ペプチド上の各アミノ酸に酸残基1個を他のアミノ酸に置換したアナログペプチドのライブラリーを用いて、T細胞応答を修飾するアナログペプチドを同定した。HLA-DRB1*0406により提示された溶連菌M12蛋白由来の15個のアミノ酸からなるペプチドに関して、DR分子に結合するアンカー残基をposition-1(P1),P4およびP6に同定した。これらの残基を置換してもT細胞クローンにアゴニズムを示してT細胞増殖反応を誘導するペプチドが多く観察されたが、T細胞レセプター(TCR)にアンタゴニズムを示すものは稀であった。いっぽう、P2,P5,P7などのTCR認識部位を置換したアナログはアゴニズムを示すものは稀で、TCRアンタゴニズムを示すものが多数観察された。さらにその中には、T細胞の容積や特定の膜蛋白の発現を増強する部分的アゴニズムを示すものが存在した。またHLA-DQ6分子により提示されたダニアレルゲンDer f I由来のペプチドに特異的なTh_2様ヒトT細胞クローンを用いて、ペプチドのN末端側よりのTCR認識部位のアミノ酸残基を置換したアナログペプチドのライブラリーに対するT細胞応答を検討した。その結果、T細胞クローンにIL-4産生や増殖応答には大きな影響を与えることなく、IFN-γの産生のみを著明に増殖するアナログを同定した。このアナログは、抗原提示細胞によるIL-12の産生増強することにより、T細胞にIFN-γの産生を増強するユニークなものであった。つまり、TCRとの結合によりHLA・ペプチド複合体を介して抗原提示細胞にIL-12産生シグナルが入り、その産生量は2つの分子の結合親和性により影響されることが明らかとなった。
