研究概要	本研究は、疾患感受性が特定のHLAクラスII(HLA-II)対立遺伝子と強く相関する自己免疫疾患において、患者の病変局所あるいは末梢血中に検出される自己免疫の標的細胞あるいは抗原提示細胞に自己反応性を示すT細胞が認識する抗原ペプチドを、同定する方法を開発することを究極の目的とする。このために遺伝子工学の手法を駆使して、多様なペプチドを提示するHLA-IIを発現する抗原提示細胞のライブラリーを作製する。まずSRαプロモーター下に連結したヒトp33インバリアント(Ii)鎖遺伝子の、CLIP領域第89‐104アミノ酸残基をコードするDNAを、任意のペプチドをコードする合成オリゴDNAと置換できるようなベクター(pCI)を作製した。pCIのCLIP部分を、M12型β溶連菌由来の非自己抗原ペプチド、あるいはヒトGAD65蛋白由来の自己抗原ペプチドに組み替えた変異Ii鎖遺伝子を作製し、HLA‐DR遺伝子と共にマウスL細胞に導入して発現させた。トランスフェクタントは、抗原ペプチドの非存在下でもHLA-DRにより提示された抗原ペプチドに特異的なT細胞クローンに増殖応答を誘導した。さらにCHO細胞トランスフェクタントを用いて、96穴プレート中の3x10^4個のCHO細胞のうち100個がTCRリガンドを発現していれば、T細胞が培養上清中に分泌するIFN-γを定量することにより、その存在を検出できることがわかった。また変異Ii鎖遺伝子を導入後間もない遺伝子発現の程度が低いトランスフェクタントを用いた場合にも、T細胞の応答を検出できた。現在、CLIP部分をランダム配列ペプチドに置換した変異Ii鎖遺伝子を作製中であり、これをCHO細胞に導入して抗原提示細胞のライブラリーとして利用する予定である。
