研究概要

移植医療において問題となっている臓器提供不足を解決するため、異種移植研究が活発に行われている。遺伝子操作により、拒絶反応をおこしにくくヒトへの移植に適した形質転換ドナー動物(ブタ)を作成することは、その臨床応用へ向けて必要不可欠である。
超急性拒絶反応を制御するために、ヒト補体制御因子(DAF,MCP,CD59)の導入に成功しているが、異種抗原であるαガラクトース(αGal)抗原の発現を抑制したブタの作出例は報告がない。遺伝子制御によるαGal抗原発現抑制方法を見出し、臨床臓器移植ドナーとしてのトランスジェニックブタ作成の有用な情報を提供することが本研究の目的である。
ブタのαGal抗原はガラクトース転移酵素(α1,3galactosyltrasferase:GT)により合成される。発現抑制にはGT遺伝子を抹消することが第一の方法として考えられ、すでにGTノックアウトマウスは2施設(アメリカ、オーストラリア)で作成されている。ブタのついては、現在ES,EG細胞は樹立されていないため、ノックアウトブタ作成は困難である。ノックアウトに代わる方法として、フコース転移酵素(α1,2fucosyltransferase:FT)などによる糖鎖抗原リモデリングが提唱されている。私どもは、FT遺伝子を導入したトランスジェニックブタ作出に成功したが、発育不良のため系統増殖には至っていない。また、in vitro transfection実験において、血管内皮細胞へFTを導入すると、細胞表層のシアル酸の減少とチューブ形成能の著明な低下がみられ、機能性糖鎖発現の変化により細胞機能の異常を来す可能性が示唆された。
別のアプローチとして、GT遺伝子をノックアウトベクターのselection markerであるネオマイシン耐性遺伝子の代わりにFT遺伝子を導入するノックインの手法を確立した。
また、アデノウイルスを用いたFT遺伝子やGTリポザイムの導入によるαGal発現抑制効果を確認した。このことは、遺伝子改変動物作成のみならず、臓器保存中に遺伝子操作を行う可能性を示している。
近い将来、ES,EG細胞や核移植技術が確立されることが想定され、私どもはブタGTゲノムのfull sequence解析を行い、targeting vectorを完成させた。同時に、GTmRNAの解析も行い、数種のsplicing variantの存在を確認した。さらに、COS細胞へのtransfection実験において、エクソン8、9を欠損したvariantはGTの酵素活性をもたず、αGalを発現させないことを明らかにした。また、これらのvariantやエクソン9のmutant cDNAによるdominant negative効果によるαGal発現抑制について検討中である。
結論として、αGal抗原発現抑制のための細胞表層糖鎖抗原リモデリングについては、それらがもたらす生理機能の変化、個体の発生、細胞の増殖・分化などへの影響はほとんど解明されておらず、いまだαGal抗原発現を十分に抑制した遺伝子改変ブタは報告されていないのが現状である。糖鎖抗原リモデリングの妥当性についての結論を急がねばならず、新たなる戦略を見出す必要に迫られている。
