研究概要

平成8年度は、マイクロスケールのSubtraction cDNAライブラリー構築のための技術的基盤を確立した。ウサギ軟骨細胞と線維芽細胞よりmRNAを抽出した。ついで軟骨細胞のmRNAよりcDNAを合成した。また、線維芽細胞より調製したmRNAをbiotin化した。これらを用いて、cDNAとmRNAの混合比率やhybridizationの条件を決定した。その結果、差し引くmRNAがcDNAの10倍-20倍あれば2回のsubtraction hybridizationで約200倍の濃縮が可能であるが、1μg以上のcDNAをsubtractionに使用しなければ実験操作中に試料の回収が困難になることが判明した。従って、より微量の試料を出発材料にしてSubtraction cDNAライブラリーを構築するためには、検索組織および対象組織より調整したmRNAをともにdscDNAに変換して、PCR増幅ののちにsubtraction hybridizationを行う方法を開発する必要がある。
そこで、微量組織より調整したcDNAをPCR法で均一に増幅するために、既存の遺伝子には見られないような配列で、まれな制限酵素切断部位を持ち、同一プライマー内あるいは異なるプライマー間で相補的な配列を持たず、かつ同じアニーリング温度のプライマーを設定できるようなアダプターおよびアンカープライマーを設計した。この、アダプターおよびアンカープライマーを結合したcDNAがPCR法で均一に増幅できることを確認した。さらに、oligo-dTを化学的にラテックスビーズに結合させたアンカープライマーを用いることで、組織のホモジネートより直接mRNAの濃縮および逆転写反応を行なうシステムを開発した。これにより、1μgのtotal RNAを出発材料としてcDNAライブラリーを構築することが可能になった。
