研究概要

本年度は、2種類の軟骨細胞特異的なSubtraction cDNA ライブラリーを作成し、それらの性能評価を行った。一つは、軟骨分化誘導したラットRMD細胞の一本鎖cDNAより未分化RMD細胞の一本鎖アンチセンスcDNAを差し引いて作成したSubtraction cDNA ライブラリーで、もう一つは、軟骨分化誘導したラットRMD細胞の二本鎖cDNAとビオチン化した未分化RMD細胞二本鎖cDNAを混合して、変性、ハイブリダイズした後、ストレプトアビジンビーズに吸着するcDNAを除くことにより作成したSubtraction cDNA ライブラリーである。両ライブラリーにおいて、軟骨細胞特異的なクローンの割合は約2割で、残りのクローンは、量的に差があるものの分化誘導したRMD細胞および未分化RMD細胞両方で発現が検出された。しかし、ライブラリー作成に必要な最低限のmRNA量は、前者と後者でそれぞれ1mgと30ngであり、後者のほうが圧倒的に少ない材料でライブラリーを作成できることがわかった。さらに、このライブラリーに含まれる遺伝子100クローンについて構造解析と発現パターンの解析を行ったところ、約8割のクローンは、細部骨格関連あるいは、細胞外基質のもので、1割は、成長因子や、代謝酵素、転写調節因子であった。残りのクローンは、これまでに報告されていないものであった。また、得られたcDNAの平均的な長さは、約550baseで、殆どのクローンの70%以上の部分は3'側の非翻訳領域を含んでいた。これらの結果より、今回検討した、Subtraction cDNA ライブラリー作成法は、微量の生体材料でも作成可能であるが、全長のクローンを得るためには、Subtraction cDNAをプローブにして、元の出発材料が由来するWhole embryoのライブラリーをスクリーニングするなどの工夫が必要であると考えられた。
