新規ドミノ反応を用いる抗癌剤カンプトテシン類の合成と原子レベルでの作用機序の解明

研究概要	カンプトテシンはトポイソメラーゼI阻害に基ずいて、強力な抗腫瘍活性を示し、その類縁体が抗がん剤として臨床において使用されている。しかしながら副作用が強く、死亡例も報告されており、より低毒性な類縁体の開発が期待されている。研究代表者は以前3,4-dihydro-1-methyl-β-carbolineと不飽和マロン酸エステルを反応させて、一行程でインドロキノリチジン体を合成する反応を開発し、この反応成績体よりカンプトテシン及び10-メトキシカンプトテシンへと誘導している。そこでまず本反応を利用してのカンプトテシン類縁体の大量合成について検討を行なったが、反応行程を短縮することは出来なかった。そこでインドロキノリチジン誘導体の効率的な合成法の開拓を目的として検討した結果、新規な分子内二重ミカエル反応を考案した。本法はインドールの2位にα,β-不飽和エステル鎖を、3位に不飽和アミド基を持つ化合物を反応基質として、酸-塩基混合剤で処理することによって、2回のミカエル反応を連続的に行ってインドロキノリチジン体を一挙に合成する方法である。最近本法を利用して、ムスカリンM2レセプターに特異的に作用するインドールアルカロイドの一種であるタカモニンの効率的合成に成功した。さらに効率的合成法の開発を目指して、キノリンの2位にアセチレン単位、3位に不飽和アミド鎖を持つ化合物を調製し、分子内ディールス・エルダー反応を検討した結果カンプトテシンの基本骨格を一挙に合成することに成功した。現在本法を活用してのカンプトテシン類縁体の合成研究を活発に行っている。
