研究概要

目的および方法:血行力学刺激に対する循環系細胞の反応(バイオメカニクス反応)は、生理的循環機能の調節に加え、循環系病態の発症、例えば、高血圧症、心肥大等とも関係が深い。本研究グループは血管細胞系を血行力学因子の受容と応答の場としてとらえ、ラット等の実験動物から摘出した脳底動脈、冠動脈および肺動脈組織や細胞等を用い、伸展や圧負荷などの刺激に対する筋原性収縮収縮反応を中心に以下の研究を行った。研究項目:1.血行力学因子の受容と応答のシグナリング特異性[(1)メカノセンサー機構の分子薬理学的研究(2)血行力学応答の細胞内/細胞・細胞間シグナリング特異性の研究]2.実験治療学的応用について[(1)新規血行力学刺激応制御薬の探索(2)モノクロタリン誘発肺動脈高血圧症病態動物を用いた研究]結果および考察:上記研究の結果、機械刺激の受容・応答機構の過程の分析が可能となった。即ち、機械応答チャネル(mechano-gated channel)タンパクのチロシンリン酸化による活性制御を介した、血管中膜平滑筋膜の脱分極とL-タイプCa^<2+>チャネルの活性化が想定された。さらに、膜電位およびCa^<2+>依存性K^+チャネルの抑制がCa^<2+>チャネルの活性化を促進することが筋原性収縮の発生に結びつくと示唆された。また、血小板由来増殖因子受容体-β鎖等の受容体型チロシンキナーゼや細胞内情報因子、例えば低分子GTPタンパク(Rho)などが機械刺激の受容・応答機構に特異的に関与する可能性が示唆された。今後の研究課題:創薬を目指す薬理学をさらに発展させるためには、生体反応に影響を与える物理的因子の存在を認識し、その定量的評価と制御に向けて新たな分野を切り開いて行くことが求められる。今後は、循環系全体を統括する血行力学受容の分子機序の提唱と血行力学受容の関わる病態の治療に向けて特にチロシンキナーゼの制御薬を足がかりにして研究を展開させたい。
