微小重力環境下における筋,骨萎縮を誘導する新規遺伝子のクローニング

研究概要	アメリカ航空宇宙局のスペースシャトルコロンビア(STS-58)に搭乗させた8週令のラット25匹と地上コントロールの同年齢ラット25匹の背柱と肋骨を入手した。その結果1)2週間の宇宙飛行では椎体骨の骨密度、骨の機械的な変化は対照群と有意な差は認められなかったが、骨組織学的検討において未熟なfibrous boneより成熟骨に置換される骨モデリングに障害が生じていることを見い出した。2)骨内微量元素をplasma atomic emission spectromertyを用いて解析したところ、宇宙飛行骨群は対照骨群に比べてFe,Zn,Mgが有意に減少していた。以上より2週間の微小重力に暴露された骨や筋肉は機能的、形態的変化を生じることを確認した。微小重力により誘導される遺伝子をクローニングするため、mRNA Diffrential Display(D.D.)法を導入した。本年度は本方法の確立のため、まずT細胞株とHTLV-1感染T細胞株のmRNAを抽出して検討し20本の異なるバンドの差を検出した。そこで宇宙飛行ラットと対照ラットの椎体骨を用いて検討を行ったが、バンド検出が得られなかった。その理由として、米国からの輸送法や保存状態に問題があり解析に必要な変性のない全長mRNAが殆ど残っていない状態であったためと思われる。そこで、われわれは傍脊柱筋の保存が良いことを利用して筋肉におけるD.D.法による解析に変更した。現在PCR法を用いてその遺伝子のスクリーニングを行っている。この解析の継続により当初予定していた実験目的が達成できると思われる。
