酵母細胞における各種蛋白質の局在に関する研究

研究概要

蛋白質の分泌過程は、分泌蛋白質が粗面小胞体上のリボソームで合成され、小胞体膜を透過し、小胞体のルーメンから輸送小胞、ゴルジ体、分泌小胞を経て細胞表面へと移動するという細胞内オルガネラを経由して、菌体外へ蛋白質が分泌する過程である。出芽酵母において、異種蛋白質を作らせ、その分泌シグナルを取り除くと、どのようになるのかを観察した。糸状菌Rhizopus niveusの菌体外酸素Aspartic proteinaseーI(RNAP-I)は成熟部分のN末端側にプレプロ配列を有している。RNAP-Iは出芽酵母においても自身のプレプロ配列により、菌体外に多量に分泌される。しかし、プロ配列を欠失した改変体(△pro)では、成熟部分の分泌が認められず、△proを菌体内に蓄積する。この△proは分泌過程において、細胞内オルガネラのどこに蓄積するかを免疫電顕法により、電子顕微鏡観察を行った。その結果、小胞体と考えられるオルガネラの顕著な発達が認められ、その内部にはelectron-denseな物質が認められた。免疫電顕法により、このelectron-denseな物質は△proの凝集体であることが明らかとなった(Yeast,13,1009-1020,(1997)発表)。現在、△proを大量に発現させた後、この蛋白質が細胞内で消失していく過程も調べている。また、小胞体シャペロン誘導に必須であるIRE遺伝子の破壊株を作成し、小胞体シャペロンとの関係も検討中である。
上記のような分泌の順行過程のみならす、逆行に関わる蛋白質の局在についても解析を進めた。ゴルジ装置から小胞体へ戻る過程に関わる遺伝子(rer2)にHAー抗原をつないで、蛋白質の局在を調べた。しかし、HAー抗体は細胞質にも反応するため、電子顕微鏡レベルでははっきりとした局在は観察されなかった。現在、mycー抗原につないだ蛋白質を作成中で、myc-抗体を使ってこの蛋白質の局在を調べる計画である。
