細菌胞子の発芽時に作用する酵素の活性化機構に関する研究

研究概要

B.cereus胞子の発芽時に現出する胞子コルテックス分解酵素の構造並びにその活性化機構の解明を試み、以下の成果を得た。
1 B.cereus由来の胞子コルテックス分解酵素は精製される酵素量が極めて少ないため、これまでその構造に関する知見を得るのが困難であった。そこで、本酵素の遺伝子クローニングを行いその全一次構造を推定したところ、本酵素はシグナルペプチドを有する分泌蛋白質であり、休眠胞子中で成熟体として存在することが示唆された。また、本酵素遺伝子は胞子形成開始約3時間後に転写が開始されること、及び転写物の大きさと転写開始点の解析から本酵素遺伝子と3'-下流に隣接するオープンリーディングフレームがオペロンを形成することを明らかにした。
2 クローン化したB.cereus胞子コルテックス分解酵素の遺伝子の組み換えによる部位特異的変異体を作製して大腸菌に発現させたところ、酵素に唯一存在するシステイン残基が活性中心ではないものの活性に重要な役割をしていることを明らかにした。
3 B.cereus胞子コルテックス分解酵素遺伝子の枯草菌における相同遺伝子をクローン化して全構造遺伝子の塩基配列を決定した。また、当該遺伝子の欠損株を作製することにより、本酵素が枯草菌胞子のL-アラニンによる発芽に特異的に応答することを明らかにした。
4 B.cereus胞子に結合する新規ズブチリシン様プロテアーゼの同定及び精製、並びに遺伝子のクローニングを行った。本酵素は、胞子発芽には直接関連しないものの、比活性が極めて高くSH修飾により失活するなどの一般のズブチリシンとは異なることを示した。
5 ショ糖脂肪酸エステルのB.cereusに対する静菌効果につき検討し、エステルの胞子結合能と静菌作用との関連を示すとともに、静菌作用の低下が胞子発芽時に遊離する酵素によるエステルの分解によることを示唆した。
