新たなビタミンDレセプター種を介する脂溶性生理活性物質の検索

研究概要

ビタミンDはカルシウム代謝に中心的な役割を果たし、更に細胞分化・増殖、また癌化などにも深く関与することが知られている。同じビタミンであるビタミンAをリガンドとするレセプターには、RAR(α、β、γ)、RXR(α、β、γ)などがあり、リガンドに対し複数のレセプターが存在する。しかしながら、ビタミンDではそのレセプターはVDR1種のみが報告されており、数多く生体内に存在するビタミンD類緑体群を考えると、VDR1種のみでは十分その生理作用を説明することができない。本研究では、VDRと類似したオ-ファンレセプターを探す過程で見出したVDRアイソフォームVDR1の機能解析と、他のビタミンD類緑体との関連を探った。その結果、VDR1は本来のビタミンDレセプター(VDR0)mRNAのスプライシングによって生じるアイソフォームであり、いわゆるイントロンがスプライトアウトされないものであった。このイントロン中には終止コドンが存在するため、VDR1タンパクはC末端に存在するリガンド結合領域を大半失っており、活性型ビタミンD[1α、25-(OH)_2D_3]では結合せず、逆にVDR0の転写促進能を阻害するいわゆるドミナンドネガティブ型のVDRであることがわかった。そこで更にビタミンD関連化合物、また未然の類似体による転写促進能を調べたところ、いずれもVDR1による転写促進能を活性化することはできなかった。一方、VDR1の標的エンハンサー配列を各種ビタミンD応答配列を用い、VDR0に比較したところ、いずれも差異が認められず、同じ配列を認識すると考えられた。
