好中球NADPHオキシダーゼの解析と活性酸素生成機構-慢性肉芽腫症の遺伝子治療の基礎的解析-

研究概要	慢性肉芽腫症の基礎的解析を行い、その遺伝子治療の基礎的データを蓄積するために本研究は行われた。そのために、スクリプス研究所Babior博士より慢性肉芽腫症患者由来のB細胞をEBウィルスの株化した細胞の提供をうけた。本細胞はNADPHオキシダーゼコンポーネントの細胞性成分であるp47phoxが欠損していることが明らかにされている。このB細胞は正常のものと違い、ホルボールで刺激してもNADPHオキシダーゼの活性化は全く観察されなかった。また欠損しているp47phoxを過剰発現させるために発現ベクターとしてEBOpLPPにp47phoxのcDNAを組み込み、この細胞に導入したところ、活性の回復が観察され、発現されたp47phoxのC末端領域の11個のセリンがリン酸化にされていることが示された。この実験結果はp47phoxはNADPHオキシダーゼ活性化に必須のものであり、p47phoxがリン酸化されることでNADPHオキシダーゼが活性化の引き金であることが明らかにされた。以上のことからリン酸化の重要性をさらに確かめるために、303と304の位置の2個のセリン残基をアラニン残基に変異を起こさせたcDNAを発現ベクターに組み込んで発現させると活性は全く見られなくなった。さらにセリンがリン酸化させたものと類似の荷電を持つグルタミン酸やアスパラギン酸残基にこの303+304を置き換えてやると、B細胞のNADPHオキシダーゼの活性は回復することが示された。以上のことから、リン酸化が活性の獲得に必須のものであることが明らかとなった。
