末梢神経再生におけるシュワン細胞と軸索の相互作用-共焦点レーザー顕微鏡による解析

研究概要

本研究課題は坐骨神経圧挫後、下肢筋の運動終板再神経支配においてシュワン細胞-軸索-細胞外物質の構造的相関が若年期(3月齢ラット)と老年期(24月齢ラット)でいかなる差異を示すかを検索することである。シュワン細胞、軸索、細胞外物質(tenascin)、接着分子(N-CAM)の各々を標識するmonoclonalまたはpolyclonal抗体を組み合わせ二重染色を行い、共焦点レーザー顕微鏡により観察した。また、これらの抗体をalpha-bungarotoxinとも組み合わせ、アセチルコリン受容器との構造的相関を解析した。老年期における特徴として前年度(平成8年度)には、次の所見を認めた。1)神経再生の早期から変性シュワン細胞が出現する、2)再生軸索は神経束から運動終板に伸びていくさい、不規則な走行や崩壊を示す、3)シナプス再構築の進展は運動終板の構成要素(終末軸索、終末シュワン細胞、アセチルコリン受容器)間で不均一である。4)N-CAM,tenascinはシュワン細胞の周縁に存在するものと考えられるが、免疫組織化学的には陽性部位の分布や形状における変化が観察される。これらの所見は軸索誘導、シナプスの再構築・維持における異常を示唆する。平成9年度にはこのような結果にもとづき論文を作成し、現在投稿中である。シナプス再構築・維持における成熟度の不均一性を更に解析するため、神経終末、終末シュワン細胞、アセチルコリン受容器の各部位のサイズおよび重複部位の面積を経時的に計測している。また、基底膜物質(laminin,proteoglycan)、髄鞘形成との関連についてもデータが必要であり、免疫組織化学的に検索している。
