動脈系化学受容器におけるアセチルコリン受容体の分子機構と形態学的解析

研究概要

1.免疫組織化学的検討:慢性低酸素下(380torr・12週)に暴露されたラット頸動脈小体は血管の著しい拡張に伴いその容積は増大し、両生類の頸動脈小体と極めて類似した形態を示した。アセチルコリン受容体(m2)に対する抗体で免疫染色をしたところ、慢性低酸素下ラット頸動脈小体におけるm2免疫陽性化学受容細胞は正常環境下に比べ多い傾向を示し、その出現頻度は正常環境下ウシガエル頸動脈小体におけるm2陽性細胞の出現頻度に近い値を示した。
2.共焦点レーザー顕微鏡による検討:アセチルコリン受容体(m2)免疫陽性部位は、化学受容細胞の膜に局在し、各種ペプチド抗体を用いて二重染色した試料では、陽性線維の近接部位により強い反応を示す傾向が認められた。
3.細胞内セカンドメッセンジャーおよびリン脂質(PI)代謝経路関連物質投与による生理応答の検討:アセチルコリン受容体から起こる反応経路を追及する為に、未固定ビブラトーム切片を作成し、Gタンパク関連物質投与下で受容体アゴニストにより刺激した。その後、サイクリックAMP(cAMP)系に関してはアデニルシクラーゼを活性化するフォルスコリンおよび膜透過性cAMPの投与を行ない細胞内小器官の運動の変化を解析した。一方、IP3系に関しては、PLCのアゴニストの投与、また、ジアニルグリセロール系に対してはTPA、OAG、H7等のCキナーゼ関連物質を用い検討した。本分析はさらに例数を重ね検討中である。
4.化学受容細胞内のカルシウム測定:未固定ビブラトーム切片を作成し、慢性低酸素状況下および正常環境下の化学受容細胞内カルシウムをfura-2を用いて顕微蛍光測光した。本分析もまた例数を重ね検討中である。
