Ca^<2+>・リン脂質結合領域(C_2)を有する新しい蛋白質の機能と作用機構

研究概要

私共の研究室では、2個のC2様領域を有する新しい蛋白質を2つ見い出し、それぞれRabphilin-3AとDoc2と命名している。これらの蛋白質は、シナプス小胞に局在し、そのC2様領域でCa^<2+>とリン脂質に結合することから、神経伝達物質の放出においてCa^<2+>センサーとして機能している可能性が高い。本年度の本研究では、Rabphilin-3AとDoc2が細胞内で神経伝達物質の放出に実際に働いていることを証明した。すなわち、私共は、Rabphilin-3AとDoc2と両者のアンチセンスや変異体の遺伝子を、ヒト成長ホルモン(GH)の遺伝子と共に、PC12細胞に強制発現させることによって、Rabphilin-3AとDoc2がCa^<2+>依存性のGHの分泌を制御することを明らかにした。さらに、Rabphilin-3Aとその変異体のリコンビナント蛋白質をヤリイカの巨大シナプスにマイクロインジェクションすることによって、Rabphilin-3Aが神経伝達物質の放出を制御することを明らかにした。一方、私共は、Rabphilin-3AがC2様領域以外のN末端側でα-アクチニンと結合し、α-アクチニンによるF-アクチニンのbundlingを促進することを明らかにした。したがって、Rabphilin-3Aは細胞骨格系をも介して神経伝達物質の放出を制御している可能性が出てきた。また、私共は、Doc2がC2様領域以外のN末端側でホルボールエステル結合領域であるCl領域を有する新しい蛋白質と結合することを見い出すと共に、この結合がホルボールエステル依存性であることを明らかにした。ホルボールエステルは神経伝達物質の放出を促進するが、Doc2とこの蛋白質との結合がその機構に作用している可能性が出てきた。このように、本年度の本研究は予想以上に進展し、当初の目的はほぼ完全に達成することができた。
