新たにクローン化した第2のFAK、CAKβ、の活性化機序とシグナル伝達機構の研究

研究概要	私達は、FAK分子族に属する第2のタンパク質チロシンキナーゼCAKβをクローン化により同定し研究している。今年度の研究により、次の事が明らかになった。1,ラット臓器・組織におけるCAKβの発現を免疫組織化学とin situ hybridizationにより検討した所、CAKβが微絨毛、繊毛、軸索に多く発現していた。2,種々の培養細胞株を用いて、CAKβの細胞内局在性を免疫細胞化学により検討した所、CAKβは上皮系細胞の細胞相互接触部位の表面膜に沿って存在する他、微小管およびミクロフィラメントと弱く結合して細胞質に多く存在した。3,CAKβに結合するタンパク質のcDNAを発現ライブラリーのscreeningにより検索し、CAKβ末端ドメインに結合するタンパク質CBP-1を同定した。CBP-1は焦点接着に局在する新規のタンパク質であり、そのNドメインでCAKβに結合する。CBP-1を免疫沈降するとCAKβが共沈した。CAKβのC末端ドメインに対する単クローン抗体で免疫染色することにより、CAKβの一部が焦点接着に存在することを明らかにした。4,CBP-1はチロシン残基のリン酸化を受けるタンパク質であった。このチロシンリン酸化は、CAKβと同様に、リゾホスファチジン酸、エンドセリンなどによる細胞の刺激に伴い亢進し、また、細胞を高浸透圧にさらすと亢進した。CAKβがCBP-1をチロシンリン酸化する可能性を検討している。また現在、CAKβの関与するシグナル路をアデノウイルスベクターに組み込んだCAKβcDNAを用いて解析している。パーソナル超遠心機は、CAKβおよびCBP-1の免疫沈降実験で抗原抗体複合体を集める目的に頻用している。
