癌特異的糖鎖抗原の合成酵素遺伝子プロモーターを用いた遺伝子治療

研究概要	1)転写調節領域の解析ヒトGM2/GD2合成酵素(β1,4-N-acetylgalactosaminyltransferase)ゲノム遺伝子を単離した。次に、転写開始点a、b、cより始まるエキソン1a、1b、1cの5´上流域の転写活性を検討した結果、エキソン1a及び1bの上流域にイントロン1を加えると転写活性は著しく上昇し、この活性の細胞特異性は本酵素遺伝子発現と相関していた。 2)GM2/GD2合成酵素遺伝子プロモーターを連結した殺細胞遺伝子発現ベクターによる殺細胞効果の特異性の検討。 GM2-GD2合成酵素遺伝子のプロモーター/エンハンサー領域(エキソン1bの上流からインロトン1領域まで)と殺細胞遺伝子(単純ヘルペスウイルスチミジンキナーゼ(HSV-TK)遺伝子)を連結したレトロウイルス発現ベクターを作製した。これを、本酵素遺伝子の高発現細胞株(AS:アストロサイトーマ)に導入し、stable transfectantを樹立した。細胞培養溶液にガンシクロビル(GCV)を添加し細胞増殖抑制効果を検討した結果、親株ではGCV:5x10^<-4>Mにおいても、増殖抑制が認められないが、stabletransfectantにおいては、GCV:5x10^<-6>Mで70%以上の増殖抑制が認められた。同様に、本酵素遺伝子の低発現細胞株MeWoに上記のレトロウイルスベクターを導入したstable transfectantでは、GCV添加による増殖抑制がほとんど認められなかった。更に、HSV-TK cDNAをプローブとした各種stable transfectantのSouthern blotを行い、同一コピー数のTK遺伝子が導入されたASおよびMeWoを選出してGCVに対する感受性を比較検討した。この場合においても、TK遺伝子導入ASにおいてGCVによる増殖抑制が認められたが、TK遺伝子導入MeWoでは、ほとんど抑制が認められなかった。
