レトロウイルス感染症に対する樹状突起細胞を用いたin vivoでの治療実験

研究概要	AIDSのモデル動物として確立されているマウスエイズ(MAIDS)を用い、樹状細胞(DC)を用いた新しい免疫療法の開発を試みた。DHは、タンパク抗原およびペプチド抗原を種々の機序で細胞内に取り込み、MHCクラスIおよびクラスII代謝経路を賦活してCD8陽性T細胞・CD4陽性T細胞の両者を活性化させる。さらに、非感作T細胞を刺激しうる極めて強力な抗原提示細胞である。我々は、MAIDSマウスの治療のため、非感染マウス骨髄細胞よりGM-CSFおよびIL-4を用いDCを誘導した。得られたDCは細胞表面にMHCクラスI・クラスII抗原・CD40・CD80・CD86などのCostimulatory抗原を強く発現していた。そこで、DCに対しin vitroにおいてウイルス感染細胞(Syngenic)を用い感作した後、マウス腹腔内に投与した。DC投与後1週でMAIDSウイルスを感染させるとともに、感作DCを週1回連続3回にわたり投与した。ウイルス感染5週で、感作DC投与マウスでは非投与ウイルス感染マウスに比し明らかに高いレベルのIL-2を産生し、CD4およびCD8陽性T細胞の活性化を認めた。しかし、抗ウイルスCTLの誘導は確認できず、B細胞の活性化を指標としたMAIDS病変の軽減も観察されなかった。さらに、脾臓ウイルス感染細胞数の現象も有意ではなかった。ウイルス感染後10週ではIL-2の産生も減少していた。こうした減少は、ウイルス抗原特異的CD4陽性T細胞の活性化がCD8陽性T細胞の活性化を凌駕した結果と考えられ、今後はDCを用いCD8陽性T細胞を特異的に活性化する方法の開発が有効と考えられた。
