寄生虫感染に伴う胸腺外分化T(NK1.1^+T)細胞の動態とその機能に関する研究

研究概要	肝類洞に見い出される胸腺外分化T細胞は、加齢、細菌内寄生細胞感染、担癌状態、及び自己免疫疾患等で著しく活性化してくる。寄生虫の感染防御においては、解析の対象とされてきたリンパ球は脾細胞を主体とする末梢リンパ球であり、これは胸腺で分化・成熟するT細胞が主体である。我々は、寄生虫感染における種々の病態で、胸腺由来のT細胞では説明できない現象が認められることから、肝T細胞を主体とする胸腺外分化T細胞の役割を解析した。 (1)マウスマラリア感染に伴う胸腺外分化T細胞の動態:マウスマラリア感染においては、肝や脾に著しいリンパ球増多が起き、特に肝の胸腺外分化T細胞が増加する。感染マウスの回復期には、γδT細胞が肝臓で著しく増加する。一方、急性期にはCD4^+細胞が回復期にはCD4^-CD8^-DN細胞が増加している。胸腺外分化T細胞に存在する自己応答性の禁止クローンは、急性期に増加した。また、感染マウスの肝リンパ球は、感染赤血球に対して著しい細胞障害活性を示した。 (2)リーシュマニア感染における胸腺外分化T細胞の動態とサイトカイン産性:L.major感染マウスにおいては、感染に伴いリンパ節や肝脾腫脹が顕著となる。そのリンパ球構成は胸腺外分化T細胞を主体とすることがわかってきた。また、この系では肝リンパ球からのIL-4を主体とするサイトカインの産性亢進が認められた。 (3)ブタ回虫感染マウスにおける胸腺外分化T細胞の動態:A.suum幼虫保蔵卵をBALB/cマウスに感染させると、幼虫の移行に伴い、原虫感染と同様に肝の胸腺外分化T細胞が一過性に活性化される。以上の結果から、寄生虫の感染防御を解析するには胸腺外分化T細胞の関与を除くことはできないことが明らかになってきた。本研究で解析が進んだ細胞内寄生であるマラリア原虫感染では、胸腺外分化T細胞の活性化が主要な免疫反応であり、しかもマラリア感染自己細胞を異常自己と認識して免疫系が働いていることが示唆された。
