HTLV-I TaxによるTNF受容体ファミリーの活性化

研究概要

TNF/TNF受容体ファミリーに属するメンバーは、TNFの他にFASやCD40など血球細胞に多彩な作用を示す。gp34は、我々が、HTLV-I発現T細胞の細胞表面の特異的分子として同定した。この分子もTNFファミリーの一員で、活性化T細胞のマーカーの一つと知られているOX40と結合する。HTLV-Iの癌化の原因遺伝子産物として考えられているTaxにより、gp34は転写レベルで発現誘導を受けることを明らかにしてきた。本研究ではTaxによるgp34の発現誘導の詳細と、OX40に及ぼす影響を調べた。gp34のプロモーター領域をクローニングしてTaxに反応するエレメントを検討した。その結果、転写開始点上流-54から-65の間のNFκB結合配列によく似たエレメントがTaxの作用に必要であり、そのエレメントにNF-κBが結合することを明らかにした。TaxがNF-κBを活性化して発現を誘導する遺伝子にはIL-2受容体α鎖などが知られているが、gp34は正常な抗原刺激以外ではT細胞に発現せずNF-κBの活性化によるものの他にTaxの制御を受けるエレメントがあるであろう。OX40の発現もHTLV-I Taxで誘導されることを証明した。
